5-Hydroxyeicosanoid dehydrogenase (5-HEDH) or more formally, nicotinamide adenine dinucleotide phosphate (NADP+)-dependent dehydrogenase, is an enzyme that metabolizes between two eicosanoid metabolites of arachidonate 5-lipoxygenase (5-LOX): 5-hydroxyeicosatetraenoic acid (5-(S)-HETE), and its 5-keto analog 5-oxo-eicosatetraenoic acid (5-oxo-ETE). It also acts in the reverse direction, metabolizing 5-oxo-ETE to 5(S)-HETE. Since 5-oxo-ETE is 30–100-fold more potent than 5(S)-HETE in stimulating various cell types, 5-HEDH is regarded as a regulator and promoter of the influence that 5-LOX and its metabolites have on cell function. Although 5-HEDH has been evaluated in a wide range of intact cells and in crude microsome preparations, it has not yet been evaluated for its structure or gene, and most studies on it have been limited to human tissues.

5-HEDH is an NADPH dehydrogenase oxidoreductase enzyme. It transfers a hydrogen cation (or hydron) H+ from 5(S)-hydroxy (i.e. 5(S)-OH) residues of its fatty acid targets to nicotinamide adenine dinucleotide phosphate+ (NADP+) to form 5-oxo (i.e. 5-O=) counterparts of its targets plus reduced NADP+, i.e. NADPH. To meet the NADPH and ATP demands in the mesophyll and bundle sheath, light needs to be harvested and shared between two distinct electron transfer chains. ATP may be produced in the bundle sheath mainly through cyclic electron flow around Photosystem I, or in the mesophyll mainly through linear electron flow, depending on the light available in the bundle sheath or in the mesophyll. The relative requirement of ATP and NADPH in each type of cell will depend on the photosynthetic subtype. The apportioning of excitation energy between the two cell types will influence the availability of ATP and NADPH in the mesophyll and bundle sheath. For instance, green light is not strongly adsorbed by mesophyll cells and can preferentially excite bundle sheath cells, or vice versa for blue light. Because bundle sheaths are surrounded by mesophyll, light harvesting in the mesophyll will reduce the light available to reach bundle sheath cells. Also, the bundle sheath size limits the amount of light that can be harvested.

Cytochrome-b5 reductase is a NADH-dependent enzyme that converts ferricytochrome from a Fe3+ form to a Fe2+ form. It contains FAD and catalyzes the reaction: In its b5-reducing capacity, this enzyme is involved in desaturation and elongation of fatty acids, cholesterol biosynthesis, and drug metabolism. This enzyme can also reduce methemoglobin to normal hemoglobin, gaining it the inaccurate synonym methemoglobin reductase. Isoforms expressed in erythrocytes (CYB5R1, CYB5R3) perform this function in vivo. Ferricyanide is another substrate in vitro.

In the evening hours of 24 June 2021, a rare, violent, and deadly long-tracked tornado struck several villages in the Hodonín and Břeclav districts of the South Moravian Region of the Czech Republic, killing six people and injuring 576 others. This tornado is the widest on record in Europe, at 3.5 kilometres (2.2 mi) maximum width. The tornado struck seven municipalities, with the worst damage in the villages of Hrušky, Moravská Nová Ves, Mikulčice and Lužice. This tornado was the strongest ever documented in modern Czech history and the deadliest European tornado since 2001. It was rated as an F4 on the Fujita scale and an IF4 on the International Fujita scale. This made it the first confirmed violent (IF4+) tornado in Europe since June 2017, when an IF4 tornado struck the village of Maloye Pes'yanovo in Russia, until the 2026 Ayyasha-Çaybeyi-Akçamezra tornado on May 3rd 2026, it was the only IF4 rated tornado to occur since the scale was introduced.

The cytochrome b5 reductase enzyme contains a typical oxidoreductase structure with a diaphorase binding domain complex for NADH and a FAD-binding domain. A three-stranded linker domain is present, as well as water-mediated hydrogen bonds, in order to biochemically connect the complex. Once cytochrome b5 reductase catalyzes electron transfer, the resulting reduced form of cytochrome b5 reduces the oxidized ferric ion of hemoglobin from Fe 3+ to Fe 2+. The protein encoded by the DLD gene comes together with another protein to form a dimer in the central metabolic pathway. Several amino acids within the catalytic pocket have been identified as important to DLD function, including R281 and N473. Although the overall fold of the human enzyme is similar to that of yeast, the human structure is different in that it has two loops that extend from the general protein structure and into the FAD binding sites when bound the NAD+ molecule, required for catalysis, is not close to the FAD moiety. However, when NADH is bound instead, it is stacked directly op top of the FAD central structure. The current hE3 structures show directly that the disease-causing mutations occur at three locations in the human enzyme: the dimer interface, the active site, and the FAD and NAD(+)-binding sites.
